Genetiska och funktionella studier av B och T cellsreceptorer – Gunilla Karlsson Hedestams grupp

Vår forskning fokuserar på adaptiva immunsvar och kvalitativa egenskaper hos B- och T-cellsrepertoarer. Ett specifik intresse i gruppen är att förstå individuella variationer i V, D och J gener och hur dessa påverkar antigen-specifika svar i kontexten av infektion, vaccination och autoimmunitet.

    Humans, like all mammals, rely on robust adaptive immune responses where secreted antibodies (immunoglobulins, Ig) play central roles in the protection against infections. Under certain conditions, antibody responses that target self antigens can develop resulting in autoimmunity, or we mount exaggerated responses to environmental antigens resulting in allergy. Why humans respond differently to immunization and why severe infections or autoimmune conditions develop in some individuals and not in others is poorly understood and likely multifactorial, but genetic components are known to be important. Despite the critical roles antibodies play in many disease settings little is known about how genetic diversity of germline-encoded Ig genes influences the protective or pathogenic potential of the expressed B cell repertoire. In an era of personalized medicine, the ability to study the Ig germline repertoire at an individual level has been a long-term goal but technical limitations have so far prevented this from becoming a reality. With the development of the IgDiscover platform in our group, it is now possible to generate personalized antibody germline databases from thousands of individuals and to study the evolution of antibody responses in selected subjects. This opens up entirely new possibilities to define the genetic diversity of the Ig loci in the human population and to study the impact of this diversity on antibody responses in health and disease.
