Seahorse XFe96 Analyser

The Seahorse XFe96 Analyser, located in Biomedicum, KI Campus Solna, can be used for a variety of applications, such as characterising disease models, identifyting drug targets, validating target effects on cellular function, and determining drug safety.

Our offer

We offer the use of a Seahorse XFe96 Analyser for metabolic characterisation of diverse cell types and ex vivo samples in a single 96-well plate.

It allows for a High-Throughput Functional Assay which can analyse baseline and stressed metabolic states in parallel. The result is a System-Level Metabolic Snapshot – integrated assessment of:

  • Mitochondrial function
  • Glycolytic activity
  • Cellular bioenergetics and dysfunction
  • Drug-induced metabolic toxicity

Key parameters of cellular metabolism in live cells are measured through Real-Time Metabolic Profiling, namely the OCR – Oxygen Consumption Rate (mitochondrial respiration) and the ECAR – Extracellular Acidification Rate (glycolysis).

The XF Real-Time ATP Rate Assay XF Real-Time ATP Rate Assay provides insights in ATP Production through quantification of mitoATP and glycoATP production rates, as well as total ATP production rate, and it enables kinetic measurement in real-time.

Available instruments

Agilent Seahorse XFe96 Extracellular Flux Analyser.

The Seahorse has been installed in a hypoxia chamber to allow for the possibility of running assays under hypoxia.

Training

One hour training for self-use of the Seahorse.

User guidelines

Researchers from KI, other academic institutions as well as biotech companies are welcome to use the Seahorse analyser.

To start using the equipment you need to login in to the iLab booking system using your institutional credentials and request a training or book a time slot.

Seahorse rules and information

  • Kindly note that only users trained by the facility staff are allowed to operate the instrument. Booking the equipment on behalf of untrained colleagues is not allowed and will be charged with a fee.
  • Please remember to always book the Seahorse through iLab.
  • Using the instrument without booking, or exceeding the booked time, will be charged with a fee.
  • In case of any issues or incidents please contact the Seahorse staff immediately via telephone (+46 (0)8 524 87938) or via email to seahorse-biomedicum@ki.se.

Please be advised that users who have not used the Seahorse analyser within the last two years must complete the training session again prior to use. This policy ensures all users maintain current knowledge of procedures and safety protocols, promoting a safe and efficient research environment for everyone.
