Om mig

  • Jag är professor i klinisk immunologi och chef för immunologiska avdelningen vid institutionen för medicinsk biokemi och biofysik,  Karolinska Institutet.

    Jag utbildade mig till läkare vid Sun Yat-sen Medical University och arbetade som läkare vid Guangzhou Respiratory Disease Research Institute (1993–1994). Jag disputerade i klinisk immunologi/immunologi vid Karolinska Institutet 1999 och genomförde postdoktoral utbildning vid Harvard Medical School (2000) och Karolinska Institutet (2001–2003). Därefter blev jag docent och gruppledare 2004 och utsågs till professor 2011.

    Jag har varit gästprofessor vid Peking University och Sun Yat-sen University Cancer Center samt gästforskare/gästprofessor vid Harvard Medical School (sabbatical), Rockefeller University (korta sabbatsperioder) och Institute of Human Biology, Roche (korta sabbatsperioder). Jag är för närvarande gästprofessor vid Tianjin Medical University Cancer Institute samt gästforskare vid Broad Institute (korta sabbatsperioder).

    Jag har handlett 14 doktorander och utbildat 24 postdoktorala forskare och har publicerat 240 vetenskapliga artiklar inom immunoglobulingenernas diversifiering, primära immunbrister, genominstabilitet, infektionssjukdomar, B-cellslymfom och cancergenetik.

  • Nuvarande projekt

    Reglering av immunoglobulin-klassbytesrekombination i mänskliga B-celler
    Detta projekt syftar till att förstå de komplexa molekylära mekanismer som är involverade i DNA-redigering, reparation och rekombination under immunoglobulin-klassbytesrekombination (CSR) och somatisk hypermutation (SHM), samt deras roller i de patofysiologiska processer som leder till immunbrist, genomisk instabilitet och cancerutveckling hos människor.

    Inducerade pluripotenta stamceller som plattform för personlig diagnos och terapi hos patienter med primär immunbrist
    Detta projekt syftar till att omprogrammera fibroblaster eller B-celler från perifert blod, hämtade från IgA-bristande (IgAD) patienter, till inducerade pluripotenta stamceller (iPS-celler) och att återdifferentiera dessa iPS-celler till antikroppsproducerande B-celler. Om detta lyckas kan metoden potentiellt ge en kurativ behandling för patienter med IgA-brist. Den kan också fungera som en metodologisk plattform för studier som syftar till cellersättningsbehandlingar hos patienter med olika andra primära immunbristsjukdomar, samt autoimmuna och neurologiska störningar kopplade till dessa tillstånd.

    Upptäckt av terapeutiska mål i B-cellslymfom med nästa generations sekvensering
    Detta projekt syftar till att identifiera potentiellt behandlingsbara molekylära mål i mogna B-cellslymfom, med fokus på diffust storcelligt B-cellslymfom, follikulärt lymfom och mantelcellslymfom, med hjälp av högkapacitativ omics-teknik för nästa generations sekvensering, såsom helgenom- och hel-exomsekvensering, bulk RNA-sekvensering, single-cell RNA-sekvensering (scRNA-seq) och spatial RNA-sekvensering. Parallellt utvecklar vi in vitro-modeller för lymfom.

    Antikroppsbehandling mot COVID-19
    Detta projekt syftar till att etablera en passiv immunterapimetod mot COVID-19, med fokus på utveckling av en mucosalt levererad IgA-baserad behandling. Strategin utvidgas också för att rikta in sig på andra luftvägs- och mag-tarminfektioner.

    Most approved antibodies against SARS-CoV-2 are ineffective against Omicron subvariants, highlighting the urgent need for alternative therapies. Secretory IgA (sIgA) is the predominant immunoglobulin type in secretions and plays a crucial role in mucosal defenses. Results from our previous VR project, "Mucosal Immunity Against SARS-CoV-2 - A Neglected Field", suggested that both dimeric and sIgA1 antibodies were superior to IgG in neutralizing SARS-CoV-2, providing protection in a mouse model through intranasal delivery. In this project, we plan to further advance the development of a broadly neutralizing IgA antibody lead candidate by generating a stable cell line with the antibody genes integrated into the genome and optimizing the production and purification processes under standard laboratory conditions. All relevant data and protocols used during cell line development and characterization will ensure compliance with regulatory standards and facilitate transfer to GMP facilities. This proof of principal proposal will help us to achieve the next goal, i.e., to further develop and commercialize the lead IgA antibody candidates as a nasal spray to block viral entry and enhance mucosal immunity against future SARS-CoV-2 variants. If successful, this approach could be extended to develop a cocktail of broadly neutralizing IgA antibodies against SARS-CoV-2 for use in a nasal spray. This will help us achieve our long-term goal of preparing for the next pandemic.
