Om mig
Jag är sedan 2012 professor i klinisk oral fysiologi och övertandläkare vid institutionen för odontologi. Sedan 2024 är jag seniorprofessor med 20% anställning och subkontrakterad PI för ett delprojekt NIH-projektet "Comprehesive functional phenotyping of trigenminal neurons innervating the temporomandibular joint (TMJ) tissues in male, female and aged mice, primates, and humans with and without TMJ disorders (TMJD)."
Jag blev tandläkare vid Karolinska Institutet 1979 och erhöll specialistkompetens i bettfysiologi 1995. Jag disputerade 1999 på en avhandling med titeln "Significance of serotonin for pain, allodynia, and hyperalgesia in the human masseter muscle" med professor Sigvard Kopp som handledare och blev docent 2004. Mellan 2004 och 2006 arbetade jag som gästforskare vid universitetet i Århus, Danmark i sammanlagt 1 år. Mellan 2009 och 2014 hade jag en halvtids klinisk forskartjänst finansierad av Vetenskapsrådet. Jag utnämndes till professor i klinisk oral fysiologi vid KI 2012.
Jag har haft flera ledningsuppdrag inom institutionen. Jag var chef för studentkliniken för vuxna mellan 2006 och 2009, avdelningschef för oral diagnostics och rehabilitation 2018–2023 och forskningsansvarig vid institutionen och därmed också med i ledningsgruppen mellan 2014 och 2022.Jag var en av de tre ansvariga för utarbetandet av Socialstyrelsens Nationella riktlinjer inom smärta och käkfunktionsstörningar. Jag är internationellt engagerad inom International Association for Dental Research (IADR) där jag var President i dess Neuroscience Group 2010. Under 2014-2016 var jag president i Swedish Pain Society, den svenska avdelningen inom International Association for the Study of Pain (IASP). Jag var samtidigt också med i den europeiska smärtorganisationens (EFIC) council. För närvarande är jag President i European Academy of Orofacial Pain and Dysfunction (EAOPD) och American Dental Association Sweden samt medlem av styrgruppen i KIs nystartade Pain Network.
Jag har publicerat ett stort antal originalartiklar, översiktsartiklar och bokkapitel.
Forskningsbeskrivning
Min forskargrupp fokuserar på att öka kunskapen om mekanismerna bakom långvarig ansiktssmärta med tonvikt på käkmuskelsmärta och könsskillnader. Forskningen är translationell och spänner från mer basvetenskapliga undersökningar på patienter och matchade smärtfria kontroller via humanexperimentella studier där smärta framkallas med olika metoder till kliniska behandlingsstudier. Vi är särskilt intresserade av potentiella perifera biomarkörer.
Det aktuella projekt jag är svensk PI för leds av professor Armen Akopian University of Texas, San Antonio och omfattar fyra PIs vid olika unversitet. Jag är den enda PI utanför USA. Projektet är i sin tur del av ett större forksningskonsortium, RE-JOIN, omfattande fem större projektgrupper och ingår i NIH sk. HEAL initiativ syftande till att hitta nya läkemedel som kan ersätta opioider vid kronisk smärta.
Forskargruppen är del av forskargruppen för orofacial neurovetenskap vid avdelningen för oral rehabilitering och samarbetar med andra forskargrupper nom KI och vid andra svenska och utländska lärosäten. Vi har också ett nära samarbete med specialistkliniken för bettfysiologi vid Eastmaninstitutet, Folktandvården AB i Stockholm.

Undervisning
Jag har haft ett stort intresse för utbildning och genom åren suttit med i kurskommittéer, linjenämnder och programnämnder. Som ämnesansvarig har var jag initiativtagare till en genomgripande förändring av undervisningen i orofacial smärta och käkfunktion, där vi gått från en traditionell undervisning med föreläsningar till "team-baserad" pedagogik. Jag var ansvarig för införandet av OSCE vid tandläkarutbildningen 2009 och för dess genomförande under flera år.
Jag har handlett 12 doktorander till disputation, 9 MSci och 39 examensarbeten för tandläkarstuderande och varit inbjuden som föreläsae både vid nationella och internationella konferenser.

- Canadian Institutes of Health Research1 april 2023 - 31 mars 2028Jaw pain is one of the most common facial pain. Among individuals affected, approximately half to two-thirds will seek professional care. Nevertheless, a large proportion of these individuals still report persistent pain 5 years later, and some are even left with long-term disabilities. Unfortunately, there is no clear evidence on how to predict or prevent the persistence of PTMD. Thus, we will conduct this longitudinal study to investigate the factors associated with the persistence of jaw pain and to identify the biomarkers. Participants will be enrolled from dental clinics in Montreal, Ottawa, Toronto, and Edmonton. They will fill out questionnaires and provide saliva during the study to measure jaw pain, and the potential factors and biomarkers associated with the persistence of jaw pain. This study will cast new light on PTMD factors that could prevent the persistence of this pain.

- National Institute of Arthritis and Musculoskeletal and Skin Diseases23 september 2022 - 31 juli 2027Abstract: Musculoskeletal (MSK) pain conditions, including temporomandibular disorders (TMD), constitute a significant and escalating global health challenge. TMD, encompassing myogenous (TMD-M) and arthrogenous (TMJD) subtypes, presents particular difficulties due to its chronic, recurrent, heterogeneous, and often debilitating nature. A primary aim of the parent project and RE-JOIN Program is to thoroughly characterize innervation, gene expression, and cellular plasticity in the temporomandibular joint (TMJ) and facial muscle tissues of patients with TMD. A comprehensive understanding of innervation and this plasticity is crucial for the development of mechanism-based and effective therapies for individuals suffering from chronic TMD pain. Over the past four years, we have identified several key challenges that hinder the comprehensive molecular characterization of muscle and TMJ tissues. Firstly, masseter muscle samples from TMD-M patients are exceedingly small (1-3 mm³), posing a methodological challenge between conventional bulk RNA sequencing and higher-resolution single-cell (scRNA-seq) approaches, which are difficult to apply to such limited material. Secondly, the inclusion of both TMD-M and TMJD patient groups enhances the translatability of the parent project but introduces additional complexity, as TMJD samples present similar constraints in tissue size, limiting the straightforward application of scRNA-seq in the cohorts. Thirdly, our experience has demonstrated that commonly used analytical approaches for bulk RNA-seq, scRNA-seq, and phenotype-to-molecular association analyses have significant limitations in terms of resolution and scalability. These limitations can be addressed using advanced computational methods. However, the increased sophistication of these methods necessitates additional computational time and dedicated personnel to keep pace with the rapidly generated datasets. These challenges will be addressed in two specific aims. Specific Aim 1 will enhance and broaden the processing of human and mouse samples through refined methods for single-cell RNA sequencing (scRNA-seq). Specific Aim 2 will employ sophisticated analytical techniques. Overall, the proposed study remains fully within the scope of parent grant UC2 AR082195. It does not alter the specific aims, involve a change in human subjects’ status, or shift the research focus to a different disease area. The primary goal of this administrative supplement is to scale human sample processing and to implement advanced analytical methods. This enhancement will improve the efficiency and impact of the parent project and directly support the goals of RE-JOIN Consortium by enabling the systematic characterization of the molecular mechanisms underlying the progression, heterogeneity, and subtypes of TMD.
