Om mig
I flera år har jag arbetat med neurovetenskaplig och farmakologisk forskning. Efter att ha avslutat min kandidatexamen 2001 påbörjade jag min forskarkarriär vid Neuroscience Scarl, Cagliari universitet, Italien. Därefter fortsatte jag med en doktorsexamen i neurovetenskap vid Cagliari universitet (2007–2010), följt av en postdoktoral tjänst vid University of Southern California, USA (2010–2012). År 2012 tilldelades jag ett Marie Curie Postdoctoral Research Fellowship för PET-BRAIN-projektet, som jag arbetade med fram till 2014. Sedan 2014, och sedan 2022 som Senior Research Specialist, arbetar jag vid avdelningen för Neurogeriatrik vid Karolinska Institutet. Jag har etablerat ett självständigt forskningsprogram med fokus på att identifiera nya målproteiner som är involverade i de biologiska mekanismerna bakom Alzheimers sjukdom. Under min karriär har jag erhållit flera nationella anslag (inklusive från Olle Engkvists Stiftelse, Alzheimerfonden, Demensfonden och Åhlén-Stiftelsen) samt internationella anslag från organisationer såsom Alzheimer’s Association.
Forskningsbeskrivning
Min forskning är inriktad på att karaktärisera nya målproteiner som är involverade i de biologiska grunderna för Alzheimers sjukdom. Specifikt studerar jag den patofysiologiska rollen av det intramembrana signalpeptidpeptidaset SPPL2b (signal peptide peptidase like 2b) i Alzheimers sjukdom. För detta ändamål använder vi den nya Alzheimers musmodellen, APP-knock-in musen AppNL-G-F, samt en muslinje som saknar SPPL2b. Min forskning utvärderar också endocannabinoidsystemets (ECS) roll vid neurologiska sjukdomar och de biologiska mekanismerna genom vilka ECS är involverat i neurodegenerativa sjukdomar som Alzheimers.

- Unraveling the Therapeutic Potential of SPPL2b in Alzheimer's DiseaseAlzhimer´s Association1 July 2024 - 30 June 2027The main question that we want to answer in this project is, how does the brain-specific intramembrane enzyme SPPL2b affect Alzheimer's disease (AD), and may it be used as a target for the creation of novel therapies? AD is characterized by an abnormal accumulation of toxic proteins (amyloid peptides) in the brain, inflammation, and progressive memory loss that affect a person’s ability to carry out daily activities. Recently, immunotherapy showed positive results in reducing plaque and slowing down cognitive decline. Still, this therapeutic approach cannot block the progression of AD and reverse memory impairment highlighting the important need in finding novel approaches to treat this disease. Our study focuses on the SPPL2b enzyme, which is involved in processing proteins involved in amyloid peptide production, inflammation, and memory process. We think that the modulation or inhibition of SPPL2b activity may help prevent the synthesis of dangerous proteins and lessen brain inflammation. We will investigate this theory by looking at the SPPL2b gene deletion results in a mouse model of AD. Additionally, we will look for SPPL2b and associated proteins in samples of human brains from various disease stages. This will enable us to comprehend the relationship between SPPL2b and AD. Additionally, we will conduct lab tests and computer simulations to look for potential SPPL2b-specific inhibitors. Our goals are to understand how SPPL2b contributes to Alzheimer's, find potential new ways to detect the disease early, and identify compounds that can block the enzyme's harmful actions. If successful, this research could lead to new drugs that slow down or halt the progression of Alzheimer's disease.
