Forskningsbeskrivning
- Oxidativ stress och kväveoxidbrist (NO) i njuren är kopplade till kardiovaskulära sjukdomar och diabeteskomplikationer. Mekanismerna är inte helt klarlagda, men ökad enzymatisk aktivitet av NADPH oxidas och onormal mitokondriefunktion kopplat till ökad produktion av reaktiva syreradikaler (inkl. superoxid) och brist på NO kan försämra kärlfunktionen och leda till försämring av njurens autoreglering och även påverka hantering av vatten och elektrolyter. Med en translationell design (d.v.s. kombination av cellstudier, djurexperimentella sjukdomsmodeller och kliniska patientstudier) syftar våra projekt till att öka kunskapen gällande bakomliggande sjukdomsmekanismer för superoxid- och NO-bildning, interaktion och påverkan på njur-, hjärtkärl- samt metabola funktioner vid hälsa och sjukdom. Olika dietära- och farmakologiska behandlingsmetoder, med syfte att dämpa oxidativ stress och öka NO produktion/signalering, undersöks och karakteriseras i experimentella sjukdomsmodeller och hos patienter med bl.a. kronisk njursjukdom, hypertoni och typ 2-diabetes.
