About me
Taher Darreh‑Shori, PhD, Docent, is a Principal Researcher at Karolinska Institutet within the Department of Neurobiology, Care Sciences and Society and the Division of Clinical Geriatrics. His work focuses on neurodegenerative diseases with particular emphasis on cholinergic mechanisms, Alzheimer’s disease, and related pathologies. [ki.se]
His research group investigates amyloid‑beta (Aβ) in relation to neuronal and non‑neuronal cholinergic signaling, develops PET tracers targeting choline acetyltransferase (ChAT) to map cholinergic networks in vivo, and explores therapeutic strategies to enhance acetylcholine production through ChAT‑potentiating ligands (CPLs).
The group also advances molecular imaging through the development and application of the synaptic PET tracer UCB‑J and the astrocytic PET tracer BU99008, an area led by Dr. Amit Kumar. [w3.ki.se]
Together, these efforts aim to improve early diagnostics and enable new treatment avenues for Alzheimer‑like dementias, ALS, and other disorders involving cholinergic degeneration.
Research
Our research focuses on cholinergic pathology in a broad spectrum of neurodegenerative diseases, including Alzheimer’s disease (AD), amyotrophic lateral sclerosis (ALS), Parkinson’s disease (PD), and atypical parkinsonian disorders such as Progressive Supranuclear Palsy (PSP) and Corticobasal Degeneration (CBD). We investigate intracellular cholinergic signaling and how mitochondrial dysfunction contributes to neuronal vulnerability, including the role of the mitochondrial serine protease OMI/HtrA2 within the apoptosis axis.
A major part of our work explores how amyloid‑beta (Aβ) peptides interact with cholinergic systems to drive pathological cascades involving synaptic disturbances, mitochondrial stress, and activation of pro‑apoptotic pathways. Increasing evidence highlights neuroinflammation and synaptic dysfunction as central contributors to early neurodegenerative processes.
In parallel, we advance cholinergic‑targeted therapeutic strategies, including the development of choline acetyltransferase–positron emission tomography (ChAT‑PET) tracers for in vivo mapping of cholinergic networks, innovative ChAT‑potentiating ligands (CPLs) to enhance endogenous acetylcholine synthesis, and our newly engineered ultra‑active ChAT nanoparticles that act as biocatalytic platforms to restore cholinergic function.
Our group also develops and applies advanced molecular imaging tools, including the synaptic PET tracer UCB‑J and the astrocytic PET tracer BU99008, to map synaptic density and astroglial activation in vivo. These imaging‑oriented subprojects are spearheaded by Dr. Amit Kumar in our laboratory.
Together, these integrated approaches aim to improve early diagnostics and enable disease‑modifying therapies across AD, ALS, PD, PSP, CBD, and other neurodegenerative disorders.

- Alzheimer's Association1 January 2016Acetylcholine is a molecule important for proper nerve cell communication. This molecule is transported between nerve cells in key areas of the brain where it helps with memory and attention. Nerve cells contain a protein called ChAT (choline-acetyltransferase) that is responsible for producing acetylycholine. In people with Alzheimer’s disease ChAT levels become reduced in brain regions affected by the disease. Changes in ChAT could therefore serve as an early biomarker of Alzheimer’s disease, but currently there is a lack of imaging techniques that can accurately “trace” ChAT levels in the living brain. Research Plan Taher Darreh-Shori, Ph.D. and colleagues have been working to develop novel compounds that can “highlight” ChAT on positron emission tomography (PET) brain scans. PET scans can be used to visualize changes over time in the levels or distribution patterns of certain molecules in the brains of people with Alzheimer’s disease. For their current studies, the researchers will further develop their PET “tracers” for ChAT. They will use research animals to determine how effectively the ChAT tracer can enter the brain when injected into the bloodstream. The researchers also plan to begin testing the safety and dosing of the ChAT tracer in a small human clinical study. Impact If successful, the results of this effort could lead to a novel PET tracer for ChAT that could improve the ability to detect Alzheimer’s disease at the earliest stages. The test could complement the other imaging modalities helping to understand the disease and improve clinical diagnosis. The study may also support the development of disease-modifying treatments by providing a new way to monitor the effectiveness of a drug.
