Jag är vikarierande prorektor på Karolinska Institutet sedan 7 mars 2016. Jag avlade läkarexamen vid Umeå Universitet 1987 och blev specialist i onkologi 1994. Under åren 1985-2007 arbetade jag kliniskt, efter 1998 parallellt med en akademisk tjänst. Merparten av det kliniska arbetet skedde på onkologiska kliniken i Umeå, där jag bl.a. under åren 1997-2001 var ansvarig för onkologisk behandling av magtarmtumörer i norra sjukvårdsregionen. Jag har också initierat och byggt upp en cancergenetisk mottagning vid Norrlands universitetssjukhus där genetisk rådgivning ges till familjer med misstänkt ärftlig cancer.

Jag disputerade 1995 vid Umeå Universitet på en avhandling om prostatacancerepidemiologi. Efter en post-doc period på Johns Hopkins i Baltimore 1996-97 återkom jag till Umeå universitet där jag blev lektor (1998) och professor (2002) i onkologi. Mitt forskningsområde är prostatacancergenetik och kliniska tillämpningar av olika genetiska markörer. Jag anställdes som professor i cancerepidemiologi vid Karolinska Institutet augusti 2005 vid Institutionen för medicinsk epidemiologi och biostatistik.

Forskningsbeskrivning

Min forskning koncentreras på att identifiera nya markörer för både risken att få prostatacancer och för prognosen för denna sjukdom när den väl diagnosticerats. Två huvudlinjer finns idag:

Identifikation och klinisk implementering av nya biomarkörer vid prostatacancer

Sedan 15 år tillbaka har min forskning varit inriktad på att identifiera medfödda genetiska variationer (SNPs) som är associerade med prostatacancer. Vår forskargrupp har tillsammans med ett internationellt konsortium, PRACTICAL, identifierat ett flertal av de idag kända 70 SNPs som associerade med prostatacancer. Dessutom har vi ett program för att identifiera nya biomarkörer (proteiner) i blodet. Vi försöker i dessa studier både hitta markörer som identifierar prostatacancer samt ännu viktigare de som är associerade med den aggressiva formen av prostatacancer.

Under 2010-2012 så inkluderade vi ca 26 000 män i STHLM2-studien (se länk längre ner) där vi systematiskt har validerat både nya och gamla biomarkörer. Som en logisk fortsättning på detta projekt så startade vi under hösten 2012 en stor randomiserad diagnostik klinisk studie STHLM3 (se länk längre ner) där vi jämför PSA med en kombinationspanel baserad på andra biomarkörer (både genetiska och proteiner).

Clinical Sequencing of Cancer in Sweden (ClinSeq)

I ClinSeq-projektet utvecklar vi nya diagnostiska tester vid cancer med hjälp av “next generation sequencing” (NGS). Vi kombinerar DNA- och RNA-sekvensering av tumörer.

1) Liquid biopsy: Från ett vanligt blodprov kan vi med avancerade metoder utvinna små DNA-bitar (ctDNA) från patientens tumör. Vi utvecklar liquid biopsy-analyser för män med spridd prostatacancer i ProBio-projektet som startade 2016.

2) Prostatacancertumörer: Från de diagnostiska biopserna som tas från prostatakörteln utvinner vi DNA och RNA. Vi utvecklar nu ett prognostisk test som ska hjälpa läkaren att rekommendera behandling för patienten.

3) ALASCCA-studien: En stor nationell randomiserad multicenterstudie av 3900 patienter med tjock-och ändtarmscancer med start 2016 där ClinSeq-projektet står för tumöranalys. PI för projektet är professor Anna Marting.
