DNA extraction at KI Biobank
DNA extracted at KI Biobank has been utilized for genotyping and NGS applications with great success. KI Biobank offers the following services on DNA extraction and reformatting:
Manual DNA extraction from 4-10 ml EDTA whole blood. The protocol is based on salt precipitation. DNA is dissolved in up to 1000 µl 10 mM Tris-HCl buffer, pH 8.0.
Automated DNA extraction. The protocol is based on magnetic bead separation. We offer DNA extraction from:
- 400 µl whole blood from 4 ml EDTA tubes. Two whole blood backup tubes are created for future DNA extraction. DNA is dissolved in 150 µl 10 mM Tris-HCl buffer, pH 8.0.
- 4 ml EDTA whole blood. DNA is dissolved in 500 µl 10 mM Tris-HCl buffer, pH 8.0.
- 4 ml saliva (Oragene®-tubes). DNA is dissolved in 500 µl 10 mM Tris-HCl buffer, pH 8.0.
Concentration and purity of DNA is measured by UV-absorbance. Sample volume used in the measurement is 3 µl.
Dilution of DNA from stock solution to microtubes (DNA normalisation) and transfer to a PCR-plate. If wanted, we also transfer DNA to other tube formats.
